Donor IVF, ICSI & IUI: How many vials of sperm do you really need to buy?

If you’re buying donor sperm for the first time, one of the most practical questions is also one of the hardest to answer: how many sperm vials do you actually need?

The answer depends on whether you’re planning IUI, IVF or ICSI, how many treatment cycles you anticipate, which type of vial your clinic accepts and whether you want to reserve sperm for future children using the same donor. However, there are some good guidelines to keep in your back pocket.

Your big question: how many vials of sperm for IVF?

For a round of donor IVF, around 1 sperm vial is generally sufficient.

IVF, or in vitro fertilization, involves fertilizing eggs with sperm in a laboratory before an embryo is transferred to the uterus. Because laboratory techniques can make efficient use of the sample, you generally need a smaller amount of sperm than you would for a treatment like IUI.

“A single vial is often sufficient for one egg-retrieval cycle,” says Larissa.

That doesn’t necessarily mean you should only purchase one vial. Requirements can vary depending on your clinic’s preferred vial type, sperm preparation method, whether ICSI will be used and how many treatment attempts you anticipate.

“Families planning for more than one child may also want to reserve additional vials upfront,” Larissa adds.

IVF can also require more than one treatment attempt, with outcomes varying significantly according to factors including age, diagnosis and treatment approach. Thinking about your potential number of cycles can therefore be useful when deciding how many vials to buy.

How many vials of sperm do you need for ICSI?

For donor ICSI, around 1 sperm vial is generally sufficient for one cycle.

ICSI, or intracytoplasmic sperm injection, is a form of IVF where a single sperm is injected directly into each mature egg during fertilization.

“ICSI allows the laboratory to use a very small number of sperm to fertilize an egg, so a single vial is often sufficient for one ICSI cycle,” explains Larissa.

Larissa says any of Cryomate’s vial types are suitable for ICSI, although your fertility clinic will determine the specific preparation and vial requirements for your treatment.

Because ICSI uses sperm so efficiently, the number of vials you need is generally driven more by the number of planned treatment cycles than by the amount of sperm required for fertilization during one cycle.

“Requirements for sperm can vary based on the clinic’s protocol, sperm preparation method, and whether additional cycles are anticipated,” Larissa says.

Finally, how many vials of sperm do you need for IUI?

For donor IUI, you’ll typically need 1–2 sperm vials per treatment.

IUI, or intrauterine insemination, involves preparing the sperm and placing it directly into the uterus around the time of ovulation.

“IUI often requires multiple treatment attempts, and because success is not guaranteed in any single cycle, families planning for IUI may want to reserve enough vials for several attempts,” explains Larissa Volz, MS, Director of Clinic Operations at Cryomate.

There’s also an important distinction between washed and unwashed sperm vials. Washed vials, sometimes called IUI vials, are prepared for use shortly after thawing and can be inserted directly into the uterus. Unwashed ICI vials require additional processing before treatment.

“Your clinic may have specific requirements for washed or unwashed vials,” Larissa says. “They can help determine the appropriate number based on your treatment plan.”

Why do these numbers vary?

The number of vials you need can vary according to your treatment type, how the sperm is prepared and how many treatment attempts you plan to make.

While the type of treatment is the first thing to consider, your clinic’s protocol, preferred vial type, sperm preparation method and whether you anticipate multiple treatment attempts can all affect how many vials you should reserve.

And if you’re planning to have future children using the same donor, that needs to be part of the calculation too.

Does age ever come into the picture?

Age doesn’t automatically determine how many sperm vials you need. 

“This again comes down to the type of treatment and how many treatment attempts you plan to do,” says Larissa.

Age can affect treatment success though, particularly as egg quality and quantity generally decline with age. This can influence fertilization, embryo development and the chance of a successful pregnancy.

And because some patients may need more treatment attempts as they get older, Larissa says they “may want to consider reserving additional vials upfront.”

When should you think about reserving vials for donor siblings?

If you hope to have future children using the same sperm donor, it’s worth thinking about sperm reservation when you purchase your first vials.

“We recommend reserving vials when purchasing for your first child — or, at the latest, once that first pregnancy is clinically confirmed,” says Larissa.

The reason is simple: donor sperm inventory is finite.

Once a donor completes their program, no additional inventory will be created. With family limits and demand, available vials can sell out within months.

“It’s also important to plan for the realities of treatment. IUI may take several attempts per child, while IVF typically requires fewer vials but still benefits from a buffer,” Larissa explains.

Okay, so how many vials are we talking?

For families hoping to have two or three full genetic siblings, Cryomate recommends considering 6–10 vials upfront.

That can feel like a significant number when you’re preparing for your first treatment, but donor sperm can be safely stored for years.

“At -196°C, metabolic activity stops completely, so properly stored sperm can retain the same viability and post-thaw recovery years later,” says Larissa.

What if you end up buying more sperm than you need?

“If you buy extra vials, they stay safely stored in our laboratory under continuous cryogenic conditions until you need them,” Larissa explains.
